RusyQuantitative tests in the other labs are like diluting the stool samples and loaded into big machines like MODULAR or COBAS. We usually send out to SGH or Parkway Lab. I am not too sure about their principle of the test.
The cassette contains antibodies specific to the Rotavirus antigen so when the antigen is present, it will bind to the antibody and then the red latex particles attached to the antibody will appear forming a red band on the cassette.
Yvonne, Xin YiThe extraction buffer is to dilute the stool sample so that the rotaviruses will be precipitated and then can be detected in the cassette. The extraction buffer contains 0.1% of sodium azide.
And to Xin Yi's question on the 50mg of the stool, referring to you pick up the solid stool sample using the wooden applicator stick and it is already 50mg when the stool is at the tip of the stick. If it is a liquid stool, 100ul of the stool is picked up and placed into the test tubes.
